This photomicrograph of a Ziehl-Neelsen stained sputum smear, revealed the presence of Mycobacterium tuberculosis bacteria. The Ziehl-Neelsen technique is used for staining acid-fast bacteria, implementing a carbol-fuchsin solution, while decolorizing in acid alcohol, and counterstaining with methylene blue. Mycobacteria have waxy coated cell walls, which prevents Gram-staining.
