This photograph depicts a close view of two slant culture tubes, each containing Difco Casitone agar supplemented with starch. The cultures were used to help identify unidentified Actinomyces species by biochemical testing. Tube A was inoculated with strain A-13-R, and Tube B with strain A-13-S. Following incubation, Gram's iodine solution was added to each tube. Tubes retaining starch, indicating an inability to hydrolyze starch, stained dark purple. The organisms in Tube A were unable to hydrolyze starch (hydrolysis negative), while those in Tube B were able to hydrolyze starch (hydrolysis positive).
