This photograph shows a close frontal view of a CDC Lombard-Dowell (LD) Presumpto Plate I quadrant plate containing LD agar lacking sodium sulfite and supplemented with esculin (1.0 g/L) and ferric citrate (0.5 g/L). Each quadrant was inoculated with an unspecified bacterium to assess esculin hydrolysis (esculinase activity). Hydrolysis of esculin produces esculetin, which reacts with ferric ions to form a dark brown to black coloration. Quadrants 1 and 2 show no color change, indicating negative results; quadrant 3 demonstrates a strong positive reaction, while quadrant 4 shows a weak positive reaction.
