This photograph shows a close frontal view of a CDC Lombard-Dowell (LD) Presumpto Plate I quadrant plate containing LD agar lacking sodium sulfite and supplemented with esculin (1.0 g/L) and ferric citrate (0.5 g/L). The quadrants were inoculated with unspecified bacteria to assess esculin hydrolysis (esculinase activity). Under Wood’s lamp illumination, hydrolysis of esculin produces esculetin, which yields a dark blue fluorescence. Quadrant 4 demonstrates a strong positive reaction, while quadrant 3 shows no fluorescence, indicating a negative result.
