This photograph shows a close frontal view of a CDC Lombard-Dowell (LD) Presumpto Plate I quadrant plate, focusing on the quadrant containing LD agar lacking sodium sulfite and supplemented with esculin (1.0 g/L) and ferric citrate (0.5 g/L). This quadrant was inoculated with an unspecified bacterium to assess esculin hydrolysis (esculinase activity). Hydrolysis of esculin produces esculetin, which reacts with ferric ions to form a dark brown to black coloration; here, a distinct darkening surrounding the bacterial growth indicates a positive reaction.
