This photograph depicted what is referred to as a two-tube nutritional study, which was arranged to test for an organism’s thiamine requirement in order to thrive. In this case, two strains of the fungal organism, Trichophyton mentagrophytes were being tested. Tubes 1a and 1b were inoculate with a downy strain, and tubes 2a and 2b were inoculated with a granular strain. The tube on the left of each pair (1a and 2a), included casein that was added to the growth medium, while the tube on the right of each pair (1b and 2b), included casein and thiamin, or vitamin B1. This test showed that these two strains of T. mentagrophytes were autotrophic for thiamine.
